Anastrozole is a nonsteroidal inhibitor of aromatase which effectively blocks estrogen synthesis in postmenopausal women and is used as therapy of estrogen receptor positive breast cancer. As first line & second line treatments, Anastrozole is indicated for the treatment of postmenopausal women with hormone receptor – positive or hormone receptor unknown locally advanced or metastatic breast cancer. It selectively binds to and reversibly inhibits aromatase, a cytochrome P-450 enzyme complex found in many tissues including those of the premenopausal ovary, liver, and breast. Aromatase catalyzes the aromatization of adrostenedione and testosterone into estrone and estradiol, (the final step in estrogen biosynthesis). In estrogen-dependent breast cancers, anastrozole may inhibit tumor growth. This slows or stops the growth of many types of breast cancer cells that need estrogen to grow. It has also been used to prevent breast cancer in those at high risk.
